dc.description.abstract | Oleaginous microorganisms are promising in the accumulation of intracellular
lipids in more than 20% of their dry biomass weight during the growth period. In
this sense, investigations were carried out evaluating the biotechnological
potential of the new strain of Penicillium spinulosum UCP1347 in accumulating
lipids using the salt-based medium, supplemented with post-frying soybean oil
and corn steep liquor, as sources of carbon and nitrogen, under different
temperatures ( 20ºC, 25ºC and 30ºC) by submerged fermentation. The highest
biomass production was observed at a temperature of 20ºC with a yield of 19.9 g
/ L, followed by 15.8 g / L at 25ºC and 14.5 g / L at 30ºC, respectively. Total lipids
were extracted from biomasses previously treated by sonication and / or
sonication and agitation, followed by the use of organic solvents, according four
methodologies based on different combinations of polarities. The results obtained
showed that P. spinulosum had a higher content of lipids for the biomass treated
by sonication followed by agitation, using the solvents chloroform: methanol (1:
1, 1: 2 and 2: 1 v / v), corresponding to highest value at 25ºC (67.80%), 20ºC
(67.02%) and 30ºC (57.80%), respectively. And, the analysis of fatty acids by
GCMS revealed a higher content of linoleic acid at all temperatures tested,
highlighting the temperature of 25ºC, followed by 20ºC and 30ºC, respectively.
The analysis of the influence of temperature on the viscosity of the lipids indicated
that at 30°C a viscosity 13.8 times higher was observed than the lipids obtained
from biomasses produced at 20 ° C (0.95cP) and 25 ° C (0 , 92Cp), respectively.
The promising technological alternatives demonstrated by the new strain of P.
spinulosum UCP1347 suggest its indication as an oleaginous fungus, with
potential for biodiesel production, but also suggests possibilities for use as
biolubricants. | eng |
